چکیده
This paper aims to explore the effect of soybean trypsin inhibitor(STI) on oxygen free radicals levels in pancreatic mitochondria of mice. 36 male KM mice were randomly divided into three groups and fed with different diets as follows: control group was fed on control diet, STI group was fed on control diet containing STI, and VC (vitamin C) group was fed on STI diet supplemented with vitamin C. Mice were sacrificed by decapitation after being fed for 3 weeks. Pancreas was quickly removed and mitochondria were prepared for determining the oxidative and antioxidative index. The results showed that, compared with mice fed with control diet, malondialdehyde (MDA) content and activity of nitric oxide synthase (NOS) in pancreatic mitochondria of mice fed with STI diet were significantly increased (P < 0.05), the content of glutathione (GSH), total antioxidant capacity (TAOC), the activities of glutathione peroxidase (GSH-Px), catalase(CAT) and superoxide dismutase (SOD) were significantly dropped. After STI diet was supplemented with antioxidant, the contents of oxidation parameters were significantly decreased (P < 0.05), but their values were still higher than those of control group; the activities of antioxidant parameters in pancreatic mitochondria were significantly increased (P < 0.05), but still lower than those of control group. These results supported that STI could cause oxidative stress in pancreatic mitochondria. In addition, vitamin C could interfere with the role of STI on the organism and effectively improve the status of

Protective effect of metformin on toxicity of butyric acid and arsenic in isolated liver mitochondria and langerhans islets in male mice: an in vitro study
Objective(s): Arsenic, a toxic metal in drinking water and butyric acid (BA) is a free fatty acid found in many foods. These two can induce oxidative stress in some tissues. The present study investigated the protective effect of metformin against toxicity induced by Arsenic (As) and BA in isolated mice liver mitochondria and pancreatic islets. Materials and Methods: In this study, liver mitoch...
متن کاملInvestigation of the Effect of Polycystic Ovary Syndrome on the Expression of the Sirtuin-3 Gene in the Ovum of the NMRI Mice
Introduction: Increasing levels of free radicals in the ova reduce the quality of their fertilization. Investigations have revealed the elevated production of tissue and serum-free radicals in women with polycystic ovary syndrome. Sirtuin-3 is located in mitochondria and plays an important role in the deactivation of free radicals. This study aimed to investigate the expression of the Sirtuin-3...
متن کاملEffects of Pre-clinical Administration of Marshmallow Extract (Althaea officinalis L.) on Certain Hepatopancreatic Enzymes in Common Carp (Cyprinus carpio)
This study evaluates the influence of marshmallow extract administration (Althaea officinalis L.) at 0.0 (control), 2.5, 5, and 10 g on the activity of certain hepatopancreatic enzymes such as α-amylase, lipase and trypsin in common carp (Cyprinus carpio) during 60 days. Feeding the fish with diets enriched with marshmallow extract had no effects on the satiety index and hepatic index in these ...
متن کاملAnalysis and optimization of nutritional set-up for murine pancreatic acinar cells.
CONTEXT Pancreatic acinar cell cultivation poses a serious problem due to limitations in the in vitro survival time despite variations of dissociation protocols, culture media and nutrient supplements. OBJECTIVE To establish a long term culture of murine pancreatic acinar cells which retain their viability, monolayer formation and responsiveness to secretagogues. In order to investigate the m...
متن کاملDevelopment of a Method for measuring Reactive Oxygen Radicals Levels In Vitro and Study the Effects of Vitamin C and E on Radical Production Reaction
Background: Free radicals and reactive oxygen species(ROS) are the most important factors in formation of oxidative stress reaction. Now, radical damage has been suggested to contribute to a wide variety of diseases such as Alzheimer, atherosclerosis and cancer. Transition metal ions in the presence of the various biomolecules produce these active compounds. The aim of this study is introducing...
